青稞的小孢子离体培养和植株再生

摘 要:

以6份青稞品系为供试材料, 研究提取和预处理过程中在提取液中添加ABA和秋水仙碱对小孢子活力的影响, 以及诱导培养基中添加不同浓度亚精胺、2,4-D和麦芽糖对愈伤组织诱导和绿苗分化产量的影响。结果表明在提取和预处理过程中提取液中添加秋水仙碱均可以提高青稞小孢子活力, 诱导培养基中添加亚精胺可以提高愈伤组织诱导和绿苗分化产量, 愈伤组织诱导培养基中2,4-D 1.5~2.0 mg•L-1和麦芽糖60~90 g•L-1浓度比较适合青稞小孢子的培养。

Abstract:

The six lines of highland barley was used to investigate the effect of abscisic acid (ABA) and colchicines in extraction buffer on the microspore viability in the process of extraction and pretreatment, and the effects of spermidine, 2,4-D and maltose concentration in induction medium on the yields of callus and green plantlets. The results showed that the microspore viability in the process of extraction and pretreatment could be improved by the addition of colchicines in extraction buffer, and the yields of callus and green plantlets could be increased by the supplement of spermidine in induction medium. The induction medium supplemented with 2,4-D 1.5–2.0 mg•L-1 and maltose 60–90 g•L-1 was suitable for highland barley microspore culture.
