冰冻切片技术在高等植物中的应用

摘 要:

冰冻切片是在低温下将组织材料冻结到一定硬度后进行切片的一种实验技术, 具有简便、高效、易操作的优点, 是植物研究中主要制片方法之一。本文对冰冻切片技术在植物组织细胞观察、组织化学研究和分子生物学研究中的应用进行了综述, 阐明了其在这些研究中的独特优势, 并对样品前处理方法、包埋剂的选择、冷冻时间、冷冻温度和切片厚度等条件进行了讨论, 旨在更好地指导冰冻切片技术在植物组织切片中的应用。

Abstract:

Cryo-sectioning is an experimental method for sectioning tissue material after it has been frozen to a certain stiffness at low temperature, which has the advantages of simplicity, efciency, and ease of operation, and is one of the main sectioning methods in plant research. In this review, we summarize the applications of cryo-sectioning in plant tissue cell observation, histochemistry research and molecular biology research, clarify its unique advantages in these studies, and discuss the conditions of sample preparation, choice of embedding agents, freezing time, freezing temperature, and section thickness, aiming to better guide the applications of cryo-sectioning in plant tissue sectioning.
