小麦籽粒抗性淀粉积累特性及淀粉合成相关酶基因表达

摘 要:

本文旨在研究小麦(Triticum aestivum)籽粒抗性淀(resistant starch, RS)积累与淀粉合成相关酶基因表达的关系, 以期为小麦营养品质改良提供一定的理论依据。以2个强筋小麦品种(‘新麦26’和‘郑麦366’)和1个黑小麦品种(‘周黑麦1号’)为材料, 研究籽粒灌浆过程中抗性淀粉积累特性及淀粉合成相关酶基因表达。结果表明, ‘新麦26’表现最高的抗性淀粉含量; 同时, 其直链淀粉含量、B型淀粉粒(粒径2~9.8 µm)表面积和数目分布百分比最高。在花后20~30 d, ‘新麦26’和‘郑麦366’具有较高的抗性淀粉增加幅度, 分别为65.6%和50.5%。籽粒发育过程中淀粉合成相关酶基因表达主要呈4种表达模式, 其中在中、后期高表达的基因可能对淀粉积累起重要的作用; ‘新麦26’在籽粒发育中、后期GBSSI的高表达以及SSI、SSIIa、SBEI和SBEIIa的较低表达可能有助于抗性淀粉含量的积累。

Resistant starch accumulation characteristics and expression analysis of genes encoding starch synthesis–related enzymes in wheat grain

Abstract:

 In order to provide references for the improvement of wheat (Triticum aestivum) nutritional quality,

the paper aims to study the relationship between the accumulation of resistant starch in wheat grains and

the expression of key enzyme genes involved in starch biosynthesis. Using two strong-gluten wheat varieties

(‘Xinmai 26’ and ‘Zhengmai 366’) and one black wheat variety (‘Zhouheimai 1’) as materials, the accumulation characteristics of resistant starch and 14 genes encoding starch synthesis–related enzyme during

grain filling were studied. The results showed that ‘Xinmai 26’ had the highest resistant starch content. At the same time, ‘Xinmai 26’ grain got the highest amylose content, and the highest distribution rate of

starch granule surface areas and the number of type B (particle size: 2~9.8 µm) starch granules. From 20

days after anthesis (DAA) to 30 DAA, ‘Xinmai 26’ and ‘Zhengmai 366’ had a big increase rate in resistant

starch content, with 65.6% and 50.5%, respectively. The expression profiles of genes encoding starch synthesis–related enzymes during grain development mainly showed four patterns, among which highly expressed in the middle and late stages may play an important role in starch accumulation. The high expres-

sion of GBSSI and the low expression of SSI, SSIIa, SBEI and SBEIIa in ‘Xinmai 26’ may contribute to the

accumulation of resistant starch content.
