海伦兜兰的无菌播种与快速繁殖

摘 要:

本文研究了海伦兜兰(Paphiopedilum helenae)的无菌播种与快速繁殖, 结果表明: 提高海伦兜兰种子萌发率较好的预处理方法是用0.1 mol•L-1 KOH处理10min; 最适合海伦兜兰种子萌发的培养基为1/4MS+0.1% (m/V)活性炭+60 g•L-1香蕉(Musa nana), 种子萌发率达69.4%; 最适合丛生芽诱导的培养基为1/2MS+0.5 mg•L-1 6-苄氨基嘌呤(6-BA)+2.0 mg•L-1萘乙酸(NAA)+0.1%活性炭+60 g•L-1香蕉; 最适合生根壮苗的培养基为MS+60 g•L-1马铃薯(Solanum tuberosum)或MS+30 g•L-1香蕉。

Abstract:

In this study, we investigated aseptic seeding and rapid propagation of Paphiopedilum helenae. The results show that the best method for pretreatment of P. helenae seeds was to soak them into 0.1 mol•L-1 KOH for 10 min; the best medium for seeds germination was 1/4MS+0.1% (m/V) activated carbon (AC)+ 60 g•L-1 banana (Musa nana), and the germinating rate was up to 69.4%; the best medium for multiple shoots induction was 1/2MS+0.5 mg•L-1 6-benzylaminopurine (6-BA)+2.0 mg•L-1 1-naphthaleneacetic acid (NAA)+0.1% AC+60 g•L-1 banana; the best medium for rooting and growth of plantlet was MS+60g•L-1 potato (Solanum tuberosum) or MS+30 g•L-1 banana.
