橡胶树铜转运蛋白HbCOPT基因克隆与表达分析

摘 要:

铜离子浓度平衡在橡胶树(Hevea brasiliensis)抗逆和橡胶生物合成中有重要作用, 铜转运蛋白又是这个平衡系统至关重要的因素。本研究中克隆得到9个铜转运蛋白(copper transporter, COPT)基因, 命名为HbCOPT1~9, 将基因推导得到的氨基酸序列进行分析, 表明HbCOPTs拥有Ctr转运蛋白特有的3个跨膜区和特异性保守氨基酸。对橡胶树中的HbCOPT1~9基因与其他物种的COPTs基因进行聚类关系分析, 结果显示, 橡胶树中的HbCOPT基因可分为3类(I、II和III)。组织特异性表达分析表明, HbCOPT4是根特异性表达基因, HbCOPT1~3HbCOPT5~8在胶乳中均有表达, 但表达量有差异。在乙烯处理条件下, HbCOPTs在12 h的表达量最高, 随后显著下降。我们的工作为进一步研究HbCOPTs在橡胶树中的生理功能和改良胶乳生产提供了依据。

Cloning and expression analysis of copper transporter genes in rubber tree (Hevea brasiliensis Müll. Arg.)

Abstract:

Copper homeostasis in rubber tree (Hevea brasiliensis) is particularly relevant to stress tolerance and rubber biosynthesis, and copper transporters play a pivotal role in copper homeostasis. In the present study, 9 genes of rubber tree copper transporter (COPT), belonging to the Ctr family of copper transporters, were cloned, and named as HbCOPT1–9. Amino acid sequence analysis shows that HbCOPTs are composed of 3 transmembrane domains and conserved amino acid residues of Ctr transporters. Phylogenetic analysis revealed the COPT genes could be divided into 3 clades (I–III). Expression studies in different tissues show that Hb-COPT4 was a root-specific-express gene, while HbCOPT1–3 and HbCOPT5–8 were detected in latex with different expression levels. Under the treatments of ethylene, HbCOPTs expression reached a maximum at 12 h after treatments, then dropped significantly. Our work provides evidence for studying physiological function of HbCOPTs in the rubber trees and modified latex production.
