摘 要：以兔眼蓝莓(Vaccinium ashei)优良品种‘粉蓝’ (‘Powderblue’)当年新发半木质化带腋芽茎段为外植体材料, 诱导腋芽萌发得到无菌种苗。选取无菌种苗幼嫩叶片, 诱导叶片直接分化产生不定芽芽丛, 然后对不定芽进行壮苗培养, 形成了再生植株。结果表明: 最佳的丛生芽诱导培养基为WPM+0.2 mg•L-1噻苯隆(TDZ)+0.1 mg•L-1萘乙酸(NAA); 最佳的壮苗培养基为WPM+0.5 mg•L-1玉米素(ZT)+0.05 mg•L-1 NAA; 壮苗后进行瓶内生根, 最佳的生根培养基为1/2WPM+0.8 mg•L-1吲哚丁酸(IBA)+0.1 g•L-1活性炭(AC)。该技术体系的建立大大缩短了组培扩繁的周期。

Abstract:Shoot segments with axillary buds from young branches of Vaccinium ashei ‘Powderblue’ were cultured in vitro. The axillary buds grew out and formed aseptic plantlets. Then young leaves of aseptic plantlets were used as explants to induce regenerating multiple shoots directly. The multiple shoots became thicker by subculturing and formed the whole plantlets finally. The results show that the optimal regeneration of multiple shoots was achieved on WPM medium supplemented with 0.2 mg•L-1 thidiazuron (TDZ) and 0.1 mg•L-1 1-naphthylacetic acid (NAA). The optimal subculture medium was WPM medium supplemented with 0.5 mg•L-1 zeatin (ZT) and 0.05 mg•L-1 NAA. The optimal rooting medium was 1/2WPM medium supplemented with 0.8 mg•L-1 3-indolebutyric acid (IBA) and 0.1 g•L-1 activated carbon (AC). The technology system could greatly shorten the propagation cycle of V. ashei ‘Powderblue’.
