药用植物北玄参高频再生体系建立

摘 要:

以北玄参的无菌苗叶片为外植体, 直接诱导不定芽并得到再生植株, 建立了北玄参叶片直接再生体系。结果表明, 北玄参种子直接用0.1% HgCl2灭菌6 min就能达到良好的消毒效果, 污染率为4.95%, 种子萌发率为81.49%; 诱导北玄参叶片再生不定芽最佳培养基为MS+6-BA 0.5 mg•L-1+NAA 0.2 mg•L-1, 诱导率可达100%; 芽增殖培养基为MS+6-BA 0.2 mg•L-1+NAA 0.2 mg•L-1, 增殖倍数为9.13; 诱导生根的培养基为1/4MS+IBA 0.2 mg•L-1, 生根率100%, 平均生根数为11.33; 再生植株移栽的成活率为86.96%。

High-Frequency Regeneration System of Medicinal Plant Scrophularia buergeriana

Abstract: The leaf from sterile seedling of Scrophularia buergeriana was acted as explants, directly induced adventitious buds and got the regenerated plant. In vitro regeneration of rapid propagation system by tissue culture was established. The results showed that immersed with 0.1% HgCl2 for 6 min was the best disinfection method for S. buergeriana seeds with the contamination rate of 4.95% and the sprout rate was 81.49%; the best medium for adventitious buds induction was MS+6-BA 0.5 mg•L-1+NAA 0.2 mg•L-1, the induction rate reached 100%; MS+6-BA 0.2 mg•L-1+NAA 0.2 mg•L-1 was the most suitable medium for buds multiplication, and multiplication ratio reached 9.13; rooting induction medium was 1/4MS+IBA 0.2 mg•L-1, and the rooting rate reached 100% and the average number of roots was 11.33. The survival rate of regenerated plantlets was 86.96% after transplanting.
