南方红豆杉试管微芽诱导培养及其紫杉醇类化合物的积累

摘 要:

以南方红豆杉幼茎段和茎尖为外植体, 以WPM为基本培养基, 单一添加不同浓度的KT、TIBA、ZT和6-BA进行芽诱 导培养。结果表明: 各自添加0.4 mg•L-1 KT、3.0 mg•L-1 TIBA、0.1 mg•L-1 ZT和0.01 mg•L-1 6-BA时的芽诱导率最高。培养50 d的试管微芽中紫杉醇和10-去乙酰基巴卡亭III (10-DABIII)的积累量比天然南方红豆杉嫩枝高2倍以上, 试管微芽中10-DABIII比紫杉醇高, 且二者均随芽诱导率的增加而增加, 当芽诱导率达最高时亦达到最高。TIBA所诱导芽短而粗壮。

The Induction and Culture of Taxus chinensis var. mairei Test-Tube MicroShoots and the Paclitaxel Compounds Accumulation in the Shoots

Abstract:

The test-tube micro-shoots were induced and cultured with Taxus chinensis var. mairei young stem segments and shoot tip in WPM medium supplemented with different concentrations of KT, TIBA, ZT, and 6-BA, respectively. The results showed that the induction rate of the bud was the highest when WPM medium supplemented with 0.4 mg·L-1 KT, 3.0 mg·L-1 TIBA, 0.1 mg·L-1 ZT, and 0.01 mg·L-1 6-BA, respectively. The accumulated volume of paclitaxel and 10-DABIII in test-tube micro-shoots at 50 days was more 2 times than the volume in the nature epicormic branch of T. Chinensis var. mairei. The content of 10-DABIII was much higher than paclitaxel in the buds. The accumulated volume of two compounds increased with increasing the bud induction rate, and the accumulation volume also reached the highest when the buds induction rate was the highest. The induced buds were short and stout when the medium supplemented with TIBA.
